Pueraria lobata (Wild.) Ohwi. (P. lobata) flowers known as ‘Kudzu flower’ contain isoflavonoids and essential oil components. They have a wide range of biological and pharmacological activities, including protective effects against non-alcoholic fatty liver disease, hyperglycemia, and hypolipidemia, anti-mutagenic effects, and benefits for weight loss. Purpose: To investigate acute toxicity, hypoglycaemic, analgesic and anti-inflammatory activities of Pulicaria incisa (Lam.) DC.Methods: Acute toxicity analysis was assessed by adopting the OECD - 423 Guidelines. The hypoglycemic effect of P. incisa infusion was investigated utilizing Oral Glucose Tolerance Test (OGTT) on orally glucose-induced hyperglycemic rats. The analgesic effect was investigated using the writhing test model, and anti-inflammatory activity was ascertained by carrageenan-induced paw oedema method.Results: The results showed that half-maximal lethal dose (LD50) values in mice were higher than 14 g/kg. The results of OGTT showed that the plant infusion produced a significant (p < 0.05) reduction in the glucose level in the blood post oral glucose administration. DPP-IV is usually found to be over expressed in many pathological conditions of Liver via disturbing immune system, lipid accumulation, ECM degradation etc. The main objective of this work was to explore the effect of methanolic extract of Pueraria tuberosa (PTME) against Alloxan induced liver damage with respect to its potential of inhibiting DPP-IV activity. PTME was prepared through continuous soxhlet extractor. Alloxan injections were given to the same age group with weight range of 80-100 g Charles foster albino male rats at the dosage of 120mg/kg bw. Rats were divided into five groups. Group 1 was given with PTME dose of 20mg/100 g bw for 7 days, Group 2 for 14 days, Group 3 for 30 days, Group 4 for 40 days and Group 5 taken as an Alloxan control. Objective : The main objective of this research was to explore the effect of the polar fraction of tubers of Pueraria tuberosa (PTWE) on DPP-IV activity. Methods : The comparison of in vitro inhibitory potential between commercially available Galvus (Vildagliptin) and PTWE was determined by measuring percent inhibition and IC 50. The enzyme kinetics were also done to reveal the nature of inhibition. In vivo study was done via the glucose tolerance test and by the measurement of increased plasma GLP-1 concentration and DPP-IV activity after glucose load. Results: PTWE has given the IC 50 value of 17.4 mg/ml and was found to be a competitive inhibitor having the Ki value of 13.11 mg/ml. Plasma GLP-1 concentration was increased and DPP-IV activity was decreased after 60 minutes of glucose load in PTWE treated rats as compared to control rats. The essential oil of leaf of P. inuloides was extracted by using hydrodistillation process and the volatile components investigated with the help of gas chromatography coupled with mass spectrometry. Diabetic rats were treated with a 400 mg/kg dose of essential oil of P. inuloides orally for 3 h (acute) or 21 days (chronic). In acute model blood glucose levels were monitored at specific intervals. In the chronic model blood samples were collected from overnight fasted diabetic rats on day 21 to estimate blood glucose level. And the body weight, serum lipid profile and activities of liver and kidney enzymes were measured. Histopathological observations of liver sections were also studied. Total seventy compounds were identified as 2-Cyclohexen-1-one, 2-methyl-5-(1-methylethyl)-, (S)-58.19%; Oxirane, 2-ethyl-2-methyl-8.25%; 1,2-Benzenedicarboxylic acid, diisooctyl ester 13.18%; n-Hexadecanoic acid 2.12% by gas chromatography coupled with mass spectrometry. Treatment of diabetic rats with 400 mg/kg of P. inuloides proved to have significant (P < 0.05) antihyperglycemic effect and have the capacity to correct the metabolic disturbances associated with diabetes. Histopathological studies showed that the essential oil of P. inuloides reinforced the healing of liver.

Antioxidant, Antidiabetic and Anti-inflammatory Potential of Phoenix pusilla Root- An Unexplored Folklore Plant.

Medicinal importance of Phoenix pusilla is unexplored. In the present study, antioxidant potential, α-amylase and α-glucosidase inhibitory effect of ethanol root extract were evaluated. Also the antiinflammatory potential was checked by two invitro methods. IC50 values of the extract against enzymes showed the ability of lowering glucose level (post prandial) at low concentration. The inhibitory action was high against α -amylase than towards α-glucosidase. Similarly the anti-inflammatory assays showed 88.62% and 67.51 % of inhibition. Thus, in vitro study revealed that the root extract have potential to act against free radicals and posess hypoglycemic and anti-inflammatory activity as determined by membrane stabilisation and proteinase inhibitory activity.

BACKGROUND: We had earlier reported that the extract of Pueraria tuberosa significantly inhibits DPP-IV enzyme, resulting in glucose tolerance response in rats. In this study, we have explored the active phytochemicals responsible for this potential. The results have been validated in both fasting and postprandial states in the plasma of normal rats and also in fasting blood and intestinal homogenates of diabetic models. METHODS: Pueraria tuberosa water extract (PTWE) was administered to normal Charles Foster rats for 35 days and to diabetic model (65 mg/kg bw) for 10 days. After treatments, oral glucose tolerance test (OGTT) and insulin was done for 90 min, and the changes in the levels of GLP-1, GIP, and DPP-IV activities were monitored in fasting and postprandial states.
